Multiple usage of the ISOLUTE multimode column for the development of a simple, rapid and cost effective method for aflatoxin analysis

Abstract
The multiple usage of Solid Phase Extraction (SPE) multimode column for sample clean-up with High Performance Liquid Chromatography (HPLC) quantification is a simple, rapid and cost effective quantitative method of aflatoxin analysis. Using the method, the recoveries of aflatoxin B1 at low (20 ppb), medium (400 ppb) and high (2000 ppb) spiked levels were 85.17 ± 4.56, 83.49 ± 1.79 and 80.13 ± 3.01% respectively. However, for aflatoxin G1 the values were 112.13 ± 3.93, 98.16 ± 1.83 and 95.55 ± 3.46% respectively. There was no significant difference (p >0.05) between each number of usage (10 times) of the column in all the three spiked levels. The overall recoveries of aflatoxin B1 and G1 using this method for the three spiked levels were 82.93 ± 3.91 and 101.94 ± 7.97% respectively. Through this technique, the cost and time taken for the clean-up process could be substantially reduced.
