Abstract

Suspension culture of Goniothalamus umbrosus represents an alternative to whole plant extraction as a source of strylpyrone derivatives (SPD). The suspension culture was established in a basal Murashige & Skoog (MS) medium supplemented with 3% sucrose with the combination of picloram (3.0 mg/litre) and benzyladenine purine (BAP) (0.1 mg/litre). Growth was measured based on the fresh and dry weights of the calli and the content of SPD produced was determined by High Performance Liquid Chromatography (HPLC). The optimum growth rate was found at incubation temperature of 25 ± 2 °C, under white light condition, pH medium 5.6 –5.8 and rotation at 100 rpm on orbital shaker. The maximum growth of cell suspension was 8.97 ± 0.08 g/litre dry weight (dw) after 30 days of incubation and SPD accumulation from 0.11– 2.14 μg/g dw sample.
